چکیده

The regulatory role of histone modifications with respect to the structure and function of chromatin is well known. Proteins and protein complexes establishing, erasing and binding these marks have been extensively studied. RNAs have only recently entered the picture of epigenetic regulation with the discovery of a vast number of long non-coding RNAs. Fast growing evidence suggests that such RNAs influence all aspects of histone modification biology. Here, we focus exclusively on the emerging functional interplay between RNAs and proteins that bind histone modifications. We discuss recent findings of reciprocally positive and negative regulations as well as summarize the current insights into the molecular mechanism directing these interactions. This article is part of a Special Issue entitled: Molecular mechanisms of histone modification function.

Long non-coding RNAs and their significance in human diseases

Protein-coding genes account for only a small fraction of the human genome and most of the genomic sequences are transcriptionally silent, but recent observations indicate significant functional elements, including non-coding protein transcripts in the human genome. Long non-coding RNAs (lncRNAs) have been defined as transcripts of >200 nucleotides without protein-coding capacity that perform t...

STU DIES ON THE BINDING OF THE ALKYLATING AGENT SULFUR MUSTARD TO CALF THYMUS CHROMATIN

In this study the effect of the alkylating agent, sulfur mustard, on calf thymus chromatin was investigated using UV/Vis spectroscopy, gel electrophoresis and thermal denaturation techniques. The results show that treatment of isolated chromatin with sulfur mustard releases histones from the core particles but does not affect histone H I and nonhistone chromosomal proteins. The content of ...

Small molecule control of chromatin remodeling.

Control of cellular transcriptional programs is based on reversible changes in chromatin conformation that affect access of the transcriptional machinery to specific gene promoters. Chromatin conformation is in turn controlled by the concerted effects of reversible, covalent modification of the DNA and histone components of chromatin, along with topographical changes in DNA-histone interactions...

Circular RNA: features, functions and their correlation with diseases especially cancer

In early 2012, the world of science saw a fascinating discovery called circular RNA as a transcription product of thousands of genes in mice and humans. These circular RNAs have recently been grouped as the encoding RNA in an independent group that their remarkable difference with other RNAs is that these RNAs are not linear, in which two ends connect with a covalent connection creating a loop-...

I-8: Role of Sperm Transcripts in Oocyte Activation, Fertilization and Development

It is well established that in addition to generating the diploid state in the zygote, the sperm also endows the oocyte with paternal centrioles, proteins and RNAs. Although the exact roles of mRNA introduced into the oocyte by the sperm are not completely understood. However, literature background, suggested that RNAs may have a vital role in embryo cleavage and further development. Therefore,...
